Conjugate adaptive Optics in Widefield Microscopy with a Novel Wavefront Sensor

Abstract:
Microscopy imaging quality is often limited by sample-induced aberrations. Adaptive optics (AO) is widely exploited to counter such aberrations. In most common applications, AO requires guide star to work and the corrected field of view is usually limited. In this talk, we show an implementation of a non-scanning conjugate adaptive optics, which exploits a novel wavefront sensor that can operate with extended light sources. As opposed to standard pupil-based AO, this implementation provides a significant field of view advantage. We also develops a technique to measure wavefront of aberrations directly utilizing light from the entire object scene, such that it neither involves the slow iterative optimization loop nor requires a localized guide star.
