SUMMARY: Conjugate Adaptive Optics in Biomedical Microscopy
DESCRIPTION:Featuring Jiang Li\n\nPart of the Departmental Seminars.\n\nDis
sertation Committee: Jerome Mertz\, Shyam Erramilli\, Pankaj Mehta\, Thoma
s Bifano\, Lei Tian (ECE)\n\nAbstract:\n\nThe quality of biomedical microsc
opy imaging is often degraded by sample-induced aberrations. Adaptive optic
s (AO) is a standard approach to counter such aberrations. In common practi
ce of AO\, an active optical correction element\, usually a deformable mirr
or (DM)\, is usually inserted in the pupil plane of the objective lens. How
ever\, the placement of the DM in a plane conjugate to a primary sample-ind
uced aberration plane can be more advantageous\, especially in situations w
here the aberrations are spatially varying and arise mainly from a dominant
layer. In this talk I will describe two novel implementations of sensor-ba
sed conjugate AO\, which do not require the use of guide stars. The first i
mplementation is applied in wide-field microscopy of transmission mode. The
wavefront sensing is based on a technique called partitioned aperture wave
front sensing that we have developed earlier. The second implementation is
in fluorescent microscopy. The wavefront sensing strategy is based on obliq
ue back-illumination. In Both implementations\, I will address the key chal
lenges of developing wavefront sensors that are capable of operating with u
ncollimated light. I will demonstrate that both implementations not only ru
n efficiently in video-rate\, but also provide large corrected field of vie
	w.\n\n![Jiang](/resources/event-image/1721/09529b0_small)
